I can shoot video with sound but people I call can’t hear me. The iPhone 6 has two microphones. Could the Front Camera secondary one be at fault? What does each microphone do? Or is there another problem? I’m stumped.

There’s three actually. One in the bottom attached to the charge port, one at the top by the front camera, and one by the rear camera. I would start by replacing the charge port.

Hi: Here’s the answer to my own question, namely, how to determine if the bottom microphone is working: Shoot a short video while saying something, first with the front camera, then with the back camera. (Recording video uses the two upper microphones, allowing you to check those.) Then, record a voice memo. (That feature uses the bottom microphone, so it will confirm whether that one’s working.)
